[Objective] Studying the functional diversity of soil microbial communities in the water-level-fluctuating zone of Dali River can provide a new theoretical basis and data support for river ecological protection from the perspective of microorganisms. [Methods] The 0—5 cm soil in the water-level fluctuating zone of the Dali River was taken as the object, the Biolog-ECO micro-plate method was used to study the average well color development(AWCD)of soil microorganisms in the water-level fluctuating zone, the utilization characteristics of different carbon sources by soil microorganisms, and the soil microbial community diversity index. [Results] The mean AWCD value of the water-level-fluctuating zone of Dali River decreases in the order: midstream>upstream>downstream. Carboxylic acids, amines, phenolic acids and polymerase are the main carbon sources used by soil microorganisms in the upstream. Amines and polymers are the main carbon sources used by soil microorganisms in the midstream and downstream. Generally speaking, amines and polymers are the main carbon sources used by soil microorganisms in the water-fluctuating-zone of Dali River. The capacity of soil microbial communities to use a single carbon source decreases in the order: upstream>midstream>downstream. Soil particles, total nitrogen, pH, and organic carbon have significant effect on the carbon source utilization capacity of microbial communities(p<0.05). [Conclusion] The functional diversity of soil microbial communities in the water-level-fluctuating zone of Dali River is different in the upper, middle, and lower reaches, and the diversity of microbial communities is affected by soil physical and chemical properties. The protection of the water-fluctuating-zone of the Yellow River should not only focus on changes in soil physical and chemical properties, but also find differences in microbial ecological functions.
