This research took three different soils in the typical root underground habitats in karst areas as the examples (TypeⅠ-dolomite level occurrence multilayer space types, Type Ⅱ-dolomite tilt occurrence multilayer space types and Type Ⅲ-dolomite vertical occurrence multilayer space types). The effect of the different types and different level on soil microbial biomass, the changes of the soil nutrient contents and soil enzyme activity were analyzed. The results showed: (1) the effect of the different plant root underground habitats and different spatial soil levels on soil microbial biomass was extremely significant(p<0.01); (2) soil microbial biomasses in three types were extremely significant different(p<0.01), it decreased with the decrease of soil level, the microbial environment of typeⅠwas poor, TypeⅡ was better, and Type Ⅲ was between the former two; (3) type Ⅰ could promote carbon cycle to a certain extent, type Ⅱ benefit was conducive to organic carbon cycle, Type Ⅲ was good for phosphorus supply and the oxidation of organic matter. The objectives of this research, are to explore the underground root habitat and its soil conditions, and provide the basis for evaluation of the soil fertility under the same conditions, and the research for vegetation restoration.
